Cryopreservation
The GMF offers several options for mouse strain cryopreservation, including:
For all services, download the Cryopreservation Service Order Form from website and submit to the GMF Administration via email: hdemanbey@mcb.harvard.edu or in Biolabs room 1062. Arrange with OAR for transfer of animal to GMF; form can be downloaded here.
Economical Solution for Freezing Embryos and Sperm GMF will freeze embryos that can be produced from one superovulation and mating experiment with up to 5 females and male mice of approximately 50 to 100 embryos in 2 to 4 straws, respectively, as well as 5 vials of sperm. Standard Embryo Cryopreservation Be sure to have 5 fertile males and at least 10 females, age three to four weeks or less from the desired strain. We will then produce approximately 250 embryos by natural mating, freeze 2-cell embryos in up to 12 straws by control rate freezing, test thaw 2 straws, implant into pseudopregnant recipients to produce live offspring. Straws will be stored in GMF cryostorage tank. Cryopreservation of Embryos produced by IVF Arrange with OAR to transfer male mice (at least two males, age 8 20 weeks, fertile). We will then produce approximately 250 embryos by IVF using fresh sperm, freeze 2-cell embryos in up to 12 straws by control rate freezing, test-thaw 2 straws, implant into pseudopregnant recipients to produce live offspring. Straws will be stored in GMF cryostorage tank. Sperm Cryopreservation Arrange with OAR to transfer male mice (at least two males, age 8 20 weeks, fertile). We will then freeze 10 vial of epididymal sperm. Straws will be stored in GMF cryostorage tank. |
