profile_image Intercellular RNA tansport: The original report of RNAi in C. elegans showed that injection of dsRNA could trigger gene-specific post-transcriptional gene silencing that could spread between tissues and cells (Fire et al., Nature 391, 806). My lab is using forward genetic analysis to identify mutations that specifically disrupt systemic RNAi, and then using the mutants and the cloned gene products to investigate how gene-specific information can be transmitted into and between cells. We have identified two proteins required for systemic spreading of RNAi, SID-1 and SID-2, whose structure, subcellular localization, and expression pattern have been informative for how double-stranded RNA can be transported into and between cells. SID-1 is transmembrane protein that appears to function as a passive pore for transport of dsRNA. Proteins homologous to SID-1 are present in mouse and human. SID-2 is a small transmembrane protein that appears to act as a dsRNA receptor for uptake of dsRNA. These studies may have a direct impact on the treatment of human genetic disease and viral infection because, although RNAi has been shown to effectively knock-down gene expression in cultured cells, a major obstacle remains efficient in vivo delivery of dsRNA into cells. Developmental Biology: Cells become different from each other by expressing different genes. To build a properly patterned embryo, the spatial and temporal expression of these different genes must be precisely controlled. My lab is investigating the mechanisms that control when and where specific transcriptional regulators are active. The early C. elegans embryo is reliant on post-transcriptional gene regulation, including control of translation, whereas transcriptional gene regulation is dominant in later embryos. Our investigations focus on PAL-1 a Caudal-like homeodomain protein that patterns posterior embryonic development. The control of PAL-1 accumulation in the C. elegans germline and early embryo is a powerful experimental system to investigate the control of translational repression and derepression. Our current work focuses on identifying and characterizing components required for post-initiation translational repression in the germline and temporally and spatially regulated de-repression in embryos. Once PAL-1 is expressed, it activates a lineage-specific network of gene activity that specifies and patterns posterior development. We are using the wealth of experimental tools available in C. elegans to take a systems level approach to understand how a single transcription factor can coordinate the specification of multiple cell fates.
