profile_image Chemical genetics describes the systematic use of small molecules to discover molecular mechanism in basic cell biology. In classical genetics, biological pathways are perturbed by mutations in genes-individual components of a pathway are altered at the level of the DNA sequences that encode them. In chemical genetics, biological pathways are perturbed by the addition of small, drug-like molecules that affect the function of individual components directly. For example, the enzyme function of a specific protein in a pathway might be inhibited when a small molecule binds to the enzyme active site. A key technology for chemical genetics is high throughput screening (HTS), in which libraries of small molecules are screened using assays, each of which is designed to report on a specific biological pathway. In HTS, tens of thousands of molecules are screened through one assay in a single day using 384-well assay plates and laboratory automation. HTS technology is also used by the biotech and pharmaceutical industries in the early stages of the drug discovery process to find "lead" compounds that might be developed into useful therapeutics. The ICCB established one of the first HTS facilities in an academic setting and I oversee its operation. I also run the ICCB Investigator-Initiated Screening Program, which provides access for research groups nationwide to the ICCB HTS Facility as well as advice on the design of HTS assay protocols. Academic researchers who screen at the ICCB are not constrained, as are scientists in pharmaceutical and biotech companies, to screen only pathways considered highly likely to produce drugs for treatment of human disease. Thus, screens at the ICCB employ a wide variety of assay methods and represent diverse biological pathways from many different organisms (e.g. viruses, parasites, bacteria, yeast, fruit flies, zebrafish, and mammals).
